    Abstract: 藉由預濃縮步驟應用於癌症的早期診斷,可更快速的進行分析並減少生物樣本的消耗,並能夠提升蛋白免疫檢測的靈敏度。本研究旨利用簡單且低成本方式,使用金奈米粒子自組裝方式沉積出奈米通道,用以達到蛋白質濃縮效果。 本旨使用不同濃度之15 nm金奈米粒子自組裝於晶片上,並使用10 μM 螢光異硫氰酸素(Fluorescein Isothiocyanate, FITC)標示之胎牛血清蛋白(Bovine Serum Albumin, BSA)進行濃縮,其結果顯示相同粒徑而不同金奈米粒子濃度其濃縮結果接近相同,而本實驗最佳合適濃度為稀釋0.5倍1.325 nM的金奈米粒子,因其搭配5 mM PBS所產生的電雙層效果最佳。不同濃度之金奈米粒子須搭配不同濃度之緩衝液才能達到最好的濃縮結果。而進行電動力濃縮至30分鐘可將提升蛋白質濃度約50倍,順利將10 μM FITC-BSA提升至0.5 mM。關鍵字:蛋白質、預濃縮、自組裝薄膜、金奈米粒子、集中排斥效應
    The protein expression in human body can be used in the detection of cancer and other diseases in the early stage. Preconcentration is an important step that increases accuracy of subsequent detection, especially for samples with extremely low concentrations. Because of overlapping electrical double layers in the nanofluidic channel, the concentration polarization effect can be generated by applying an electric field. In this study, the preconcentration chip for proteins was fabricated using simple standard soft lithography with a polydimethylsiloxane (PDMS) replica. Using various concentration gold nanoparticles (1.325、2.65 and 26.5 nM) deposited on glass substrate by self-assembly. The results showed apparently that all case using 15 nm gold nanoparticles for SAM achieved protein concentration successfully. The experimental results indicate that gold nanoparticles concentration do not affect concentration results. Over concentration gold nanoparticles (26.5 nM) have regular pattern and compact structure. A protein sample with an concentration of 10 μM was concentrated to 50-fold in 30 min by using the proposed chip.Keywords: Protein, Preconcentration, Self-assembled monolayer, Gold nanoparticles, Exclusion-Enrichment Effect
